Nikki Rene Glaser was born on June 1, 1984, in Cincinnati, Ohio, to Julie E. and Edward J. Glaser. She has a younger sister named Lauren. Glaser was raised in a Catholic family of German and Irish descent. The family later moved to the St. Louis suburb of Kirkwood, Missouri, where she spent most of her childhood.

Glaser attended Kirkwood High School, graduating in 2002. Following high school, she briefly attended the University of Colorado Boulder before transferring to the University of Kansas.
